    The topic "Nutrition and anti-oxidant functions" comes within the research context on nutritional quality control of farmed fish for human consumption. Indeed, the nutritional qualities and health-enhancing properties of fish are mainly due to the high level of polyunsatured fatty acids n-3. However, due to the presence of double bonds, polyunsaturated fatty acids are prone to oxidation. Besides the decrease of nutritional and organoleptic qualities, lipid peroxidation leads to the higher occurence of harmful compounds which have to be maintained at a low level.
    The research project aims to study the nutritional regulation of anti-oxidant functions in fish from early developmental stages in order to preserve polyunsaturated fatty acids in flesh and limit stress oxidative-associated pathologies (for instance: malformations in fish larvae, Figure 1B).

These anti-oxidant functions consist mainly of enzymes (superoxyde dismutase, catalase and glutathione peroxidase, reductase and transferase) and vitamins. The objective of the ongoing studies is:

  • to test the anti-oxidant role of selected vitamins such as vitamin A in fish broodstock and larval nutrition,
  • to study the nutritional regulation of anti-oxidant enzymes during early life stages in fish larvae fed diets with pro-oxidant ingredients (oxidized lipid in controlled conditions or highly unsaturated lipid) and/or anti-oxidant ingredients (vitamins E and A, astaxanthin).

We study the metabolism of vitamins by HPLC and the control of anti-oxidant enzymes by activity measurements and by quantitative RT-PCR for expression analyses of genes encoding anti-oxidant enzymes. These analyses are completed by the localization of gene expression performed by in situ hybridization on early life stages. The impact of these parameters on the larval development is evaluated (absence of any malformations).

The first results indicate differences in stress oxidant sensitivity among fish species and developmental stages. For instance rainbow trout fry fed oxidized lipid display an increase of anti-oxidant enzyme activity which allow to avoid oxidative stress whereas in the same situation, the absence of control of anti-oxidant enzymes in Siberian sturgeon larvae leads to oxidative stress which results in an increase of lipid peroxidation products in flesh and higher incidence of malformed fish (Figure 1B).
