THEMES DE RECHERCHE

Au sein de l’équipe GFDF, une approche de génomique fonctionnelle est développée sur la variété naine de tomate Micro-Tom en tant qu’espèce modèle pour les fruits charnus. Commencé en 2001, une collection de mutants fortement mutagénéisée a été réalisée pour obtenir plusieurs mutations ponctuelles dans chacun des 35000 gènes de la tomate.  L’ADN des 8500 mutants a été extrait et utilisé dans l’approche de génétique inverse, le TILLING. Le phénotypage d’une partie de la collection (3500 familles de mutants, 42000 plantes) a été réalisé sur la base de 168 critères phénotypiques différents. L’ensemble représente 26000 annotations et 4000 photographies qui alimentent la base de données MMDB (Micro-Tom Mutant Data Base) accessible sur le site INRA Aquitaine. Actuellement, l’on tend vers la valorisation de la collection de mutants par différents axes : (1) la diffusion de graines soit sur la base du phénotype décrit dans la base soit après des résultats issus du TILLING, (2) l’étude de mutants de couleur du fruit dans le cadre d’un projet européen TomQML, (3) le développement d’une approche de cartographie grossière avec le croisement avec la variété M82 en collaboration avec le Kazusa DNA Research Institute, (4) le projet de développement d’une autre approche de génétique directe par l’identification chez des mutants d’intérêt de la mutation responsable du phénotype par WGS (Whole Genome Sequencing).
